| 要旨本文 | High repeat content and heterozygous conifer genomes remain challenging to assemble. Here, we report a haplotype-resolved chromosome-level genome of Sakhalin fir (Abies sachalinensis), one of the major species for silviculture and ecosystem in northern Japan. Using HiFi sequencing and RNA-seq, we assembled two haplotype genomes (HapA 15.91Gbp, HapB 15.78Gbp) and annotated complete protein-coding genes (HapA 46,145, HapB 46,116). The assemblies show high contiguity and completeness (BUSCO: HapA 90.5%, HapB 90.5%), and reveal structural differences between haplotypes. High repeat content (HapA 82.77%, HapB 82.92%) was detected in both haplotypes. This haplotype-resolved chromosome-level genome provides an allelic view of gene content, structural variation, and repeat content. The results can be applied in local adaptation and genome evolution in Abies and related conifers. |
